作用機(jī)制:
潮霉素B是由吸水鏈霉菌(Streptomyces hygroscopicus)代謝產(chǎn)生的一種氨基糖苷類抗生素,通過抑制蛋白質(zhì)合成而殺死細(xì)菌、真菌和高等真核細(xì)胞。潮霉素B通過干擾70S核糖體易位和誘導(dǎo)對mRNA模板的錯讀而抑制蛋白合成。目前常用于篩選和維持培養(yǎng)含潮霉素抗性基因的原核或真核細(xì)胞。
抗性基因:
對潮霉素B的抗性源自編碼潮霉素B磷酸轉(zhuǎn)移酶(Hph)的基因。至今發(fā)現(xiàn)兩種來源:
► Streptomyces hygroscopicus產(chǎn)生的Hph抗性基因;
► Escherichia coli,Klebsiella pneumoniae內(nèi)質(zhì)粒攜帶的Hph抗性基因(常用);
生物應(yīng)用:
1) 篩選和維持培養(yǎng)穩(wěn)定轉(zhuǎn)染Hph+載體的原核細(xì)胞或真核細(xì)胞(植物細(xì)胞和哺乳動物細(xì)胞);
2) 由于作用模式的差異,常與G418 (GeneticinTM),Zeocin™ 和blasticidin S聯(lián)合使用,篩選穩(wěn)定轉(zhuǎn)染兩個不同載體的細(xì)胞株;

應(yīng)用濃度:
潮霉素B用來篩選穩(wěn)轉(zhuǎn)株的工作濃度需要根據(jù)細(xì)胞類型,培養(yǎng)基,生長條件和細(xì)胞代謝率而變化。推薦使用濃度為50-1000 μg/mL,最佳濃度需要?dú)缜來確定。
哺乳動物細(xì)胞·200-500 μg/mL;細(xì)菌/植物細(xì)胞·100-300 μg/mL;真菌·300-1000 μg/mL;
產(chǎn)品使用:
使用一:殺滅曲線確定最佳殺死濃度(僅作參考,因個人檢測體系而異)
(1)往組織培養(yǎng)級的96孔板內(nèi)加入未轉(zhuǎn)染細(xì)胞,細(xì)胞密度約50-200細(xì)胞/孔;細(xì)胞貼壁之后,往每孔加入含不同濃度(如50-1000μg/mL,至少5個濃度)潮霉素B的200μL新鮮培養(yǎng)基;
(2)37℃,5% CO2培養(yǎng)箱孵育細(xì)胞10-14天;
(3)培養(yǎng)5-7天后更換新的培養(yǎng)液,培養(yǎng)液內(nèi)含有相應(yīng)濃度的潮霉素B;
(4)10-14天后使用細(xì)胞增殖方法如MTT,CCK-8等評估細(xì)胞活力;也可以通過檢測細(xì)胞克隆數(shù)或百分比匯合率來確定毒性效應(yīng)。一般選擇在10-14天能夠殺死所有細(xì)胞的最小濃度為最佳篩選濃度。
使用二:篩選穩(wěn)定轉(zhuǎn)染細(xì)胞
(1) 對于貼壁細(xì)胞,直接吸去60mm培養(yǎng)皿內(nèi)的轉(zhuǎn)染細(xì)胞培養(yǎng)基,然后加入含有潮霉素B的新鮮培養(yǎng)基5-6ml;對于懸浮細(xì)胞,無菌條件250x g,離心10min除去舊的轉(zhuǎn)染細(xì)胞培養(yǎng)基,然后懸浮在約5ml的含潮霉素B的新鮮培養(yǎng)基;
(2) 5-7天后,如上方法更換含有潮霉素B的新鮮培養(yǎng)基;
(3) 再孵育細(xì)胞5-7天;
(4) 10-14天孵育后,培養(yǎng)體系中只含有能夠表達(dá)潮霉素B抗性表型的活細(xì)胞。因此篩選之后如步驟一,更換不含潮霉素B的新鮮培養(yǎng)基。
