按理化性質和生物學功能,可將其分為IgM、IgG、IgA、IgE、IgD五類。IgM抗體是免疫應答中首先分泌的抗體。它們在與抗原結合后啟動補體的級聯(lián)反應。它們還把入侵者相互連接起來,聚成一堆便于巨噬細胞的吞噬;IgG抗體激活補體,中和多種毒素。IgG持續(xù)的時間長,是唯一能在母親妊娠期穿過胎盤保護胎兒的抗體。他們還從乳腺分泌進入初乳,使新生兒得到保護;IgA抗體進入身體的黏膜表面,包括呼吸、消化、生殖等管道的黏膜,中和感染因子。還可以通過母乳的初乳把這種抗體輸送到新生兒的消化道黏膜中,是在母乳中含量最多,最為重要的一類抗體;IgE抗體的尾部與嗜堿細胞、肥大細胞的細胞膜結合。當抗體與抗原結合后,嗜堿細胞與肥大細胞釋放組織胺一類物質促進炎癥的發(fā)展。這也是引發(fā)速發(fā)型過敏反應的抗體;IgD抗體的作用還不太清楚。它們主要出現(xiàn)在成熟的B淋巴細胞表面上,可能與B細胞的分化有關。(IgD于1995年從人骨髓瘤蛋白中發(fā)現(xiàn),分子量為175kD,主要由扁桃體、脾等處漿細胞產生,人血清中IgD濃度為3~40μg/ml,不到血清總Ig的1%,在個體發(fā)育中合成較晚。IgD鉸鏈區(qū)很長,且對蛋白酶水解敏感,因此IgD半衰期很短,僅2.8天。血清中IgD確切的免疫功能尚不清楚。在B細胞分化到成熟B細胞階段,除了表達SmIgD,抗原刺激后表現(xiàn)為免疫耐受。成熟B細胞活化后或者活化后或者變成記憶B細胞時,SmIgD逐漸消失。
